Affymetrix GeneChip Resource at the HKU Centre for Genomic Sciences

The Affymetrix GeneChip Resource at HKU Centre for Genomic Sciences is a core service facility dedicated to provide RNA gene expression profiling and whole genome DNA Genotyping services. The Resource is open to both HKU and non-HKU researchers.

The GeneChip technology platform consists of high-density microarrays and tools to help process and analyze these arrays, including standardized assays and reagents, instrumentation, data management and analysis tools.

Established since July 2003, our platform is now equipped with two hybridization ovens, two fluidic stations and a 7G scanner with GeneChip autoloader controlled by a dedicated computer workstation. This set-up allows a high-throughput round-the-clock processing of GeneChips.

Our platform specialists are experienced in handling and processing various samples and GeneChips. Quality control is applied at every recommended step. Affymetix has visited the CGS microarray platform for multiple on-site trainings, during which the laboratory protocols and data quality have been fully approved.


Affymetrix Technology Overview

Affymetrix GeneChip microarrays consist of small DNA fragments (probes) chemically synthesized at specific locations on a coated quartz surface.  Different combinations of probe design enable a wide range of applications on a whole-genome scale, including gene- and exon-level expression analysis, novel transcript discovery, genotyping, and resequencing.  Combined with chromatin immunoprecipitation, genome-wide identification of transcription factors and their respective binding sites can also be studied.

A typical Affymetrix GeneChip experiment includes the following steps:

  • Preparation of labeled-targets from RNA/DNA isolated from specimens
  • Hybridization of targets to arrays
  • Stringent washes, staining and scanning of arrays
  • Analysis of scanned images and data extraction

A broad survey of gene expression level and genotypes of the genome is possible by a single hybridization of the labeled targets to an array.
