Abstract:

Bifidobacterium animalis subsp. lactis, a probiotic bacterium that naturally colonizes the human gastrointestinal tract (GIT), is of essential importance in promoting infants' health and has been broadly used as a yogurt additive. B. animalis subsp. lactis V9 was isolated from the feces of a healthy Inner Mongolian infant in China. This strain possesses some good probiotic properties such as high resistance to acidic pH and bile salt. The approximately 2.0 Mb circular chromosome genome was sequenced with more than 400x coverage by next-generation sequencing platform SOLiDâ„¢ using a mate-paired library of about 3 kb insert-size and 2x25 bp read length. Up to date, we have gotten the 18 contigs V9 genome draft through the combination of SOLiDâ„¢ mapping algorithm, BLAST, Phrap and Velvet de novo assembler. A total of 1567 genes (61.38% GC) were identified from the n V9 genome draft (60.47% GC). Most of the contig ends are flanked by repeat sequences and we are currently performing PCR amplification and traditional Sanger sequencing of these regions in order to close gaps.
