Abstract:

With the advent and availability of whole genome arrays and Next Generation Sequencing technologies, the current challenge isn’t necessarily with the identification of new biomarkers, but in the screening and validation -- confirming that the biomarkers are accurate and predictive. For established biomarker panels, clinical validation, be prospectively and retrospectively, will require large patient cohorts in order to achieve statistical significance of the sensitivity, specificity, positive predictive value and negative predictive value. New technologies were developed for biomarker screening and validation utilizing TaqMan® Real Time PCR arrays with different reaction densities to accommodate varying cohort sizes. It enables interrogation a large number of nucleic acid samples against a large number of assays in flexible, configurable formats. By altering the number of assays or the number of samples the researcher can easily customize the arrays to meet their changing biomarker needs. Applications and performance of these arrays will be discussed including digital PCR arrays for absolute quantification of nucleic acid samples and castPCR for detecting rare mutant alleles for up to 1 in 10,000,000.
