Abstract:

Antisense techniques have been employed for over 30 years to suppress expression of target RNAs. More recently, RNA interference technology have emerged as a new means of gene regulation. Small interfering RNAs (siRNAs) have become popular tools in molecular biology to perform RNA knockout experiments. Dicer-substrate RNAs (DsiRNAs) is a new class of siRNA molecules developed by Dr. Rossi and IDT. DsiRNAs are chemically synthesized 27-mer duplex RNAs, which show increased potency in RNAi compared to traditional siRNAs. Traditional 21-mer siRNAs are designed to mimic Dicer products and therefore bypass interaction with the enzyme Dicer. Dicer has been recently shown to be a component of RISC and involved with entry of the siRNA duplex into RISC. DsiRNAs are designed to be optimally processed by Dicer and show increased potency by engaging this natural processing pathway. New design rules specific to DsiRNAs have been developed and are available only from IDT.
