Abstract:

The field of miRNA represents a new dimension to global regulatory networks that is important for cell physiology. miRNAs are small non-coding RNAs predicted to regulate the expression of up to 30% of all protein-coding genes by binding to sites within the 3' UTR of mRNAs. The study of miRNA requires special approaches to sensitively and accurately detect such small molecules. Exiqon has developed several technological platforms based on Locked Nucleic Acid (LNATM) to profile miRNAs and uncover their functions. LNATM comprises a unique class of RNA analogues. The exceptional thermal stabilities and high hybridization affinity obtained by incorporating LNATM in oligonucleotides enables design of sensitive and specific tools for miRNA expression analysis. We will discuss use of these LNATM-based tools for sensitive and accurate discovery, profiling, validation, localization and also knockdown of miRNAs. Expression profiling of miRNAs has been demonstrated to be a more accurate method of classifying cancer subtypes than expression profiles of protein coding genes. Therefore miRNA expression profiles have great potential to be used as diagnostic and prognostic markers for cancer and other diseases.
