MIQE guidelines the assessment of total RNA integrity, the normalisation strategy and data analysis are essential steps to receive meaningful mRNA and microRNA expression profiling results. The talk will cover the “pros & cons” performing relative mRNA quantification strategies in RT-qPCR and will illustrate the evolutional developments made in the past decade. The discussion turns into practical considerations with focus on standardisation according to the MIQE guidelines.

An overview of the latest transcriptomics based technologies in biomarker discovery after doping will be presented. It highlights recent gene expression results on mRNA and microRNA level derived from quantitative RT-PCR and next generation sequencing (NGS). To verify if these significant changes in the gene expression pattern could act as first valid biomarkers advanced bio-statistical methods were applied. Principal Components Analysis (PCA) and Hierarchical Clustering (HCA) were successfully used to demonstrate the potential of the transcriptomic approach for the development of a new screening system and to introduce new gene expression biomarkers.
