Abstract:

As the major approach for proteomics, classical mass spectrometry (MS) faces new challenges, such as throughput limitation for large-scale protein identification, complex matrix interference for quantification, and ineffective fragmentation for PTM characterization. Recent progress in Orbitrap MS technologies permits new breakthrough in proteomics. Identification of more than 4000 proteins within one hour is realized for large-scale proteomic analysis by massive parallelization. Comprehensive characterization of intact glycopeptides with peptide sequence, site localization as well as glycan structure elucidation is achieved using latest fragmentation and MSn technologies. Quantification reproducibility and accuracy is significantly improved with novel MS workflows, including synchronous precursor selection (SPS), parallel reaction monitoring (PRM), multiplexing acquisition (MSX), and various data independent acquisition (DIA) approaches. This seminar will introduce the most recent advances in proteomic analyses, and discuss the possible development these new technologies will bring to proteomics.
